position. The ZMBP comprises 17 research groups and central facilities with nearly 200 staff members. The successful candidate will be expected to establish a competitive research program in the scientific area of

Molecular Plant Biology

and to interact with the research groups of the ZMBP. Collaborations with other groups within the Science Faculty of the University and the MPI for Developmental Biology are also welcome.

The position is vacant immediately and initially limited to 6 years. Salary and social benefits are according to scale E13 of the German Civil Service Regulations.

The Center provides lab space with full equipment, funding for staff, start-up money, an independent annual budget and shared equipment funds.

The research group will benefit from the ZMBP central service units (administration, plant transformation, cytometry/FACS, greenhouse facilities, analytics, microscopy, IT support).

Research: Our group at the ZMBP Tübingen is interested in the role of protein homeostasis networks in plant-microbe interactions. In our interdisciplinary group, using cell biological, biochemical and genetic approaches, we are studying the intimate interplay between protein synthesis, trafficking and degradation and how these pathways co-ordinate their functions to regulate plant immunity.

In the ERC funded project DIVERSIPHAGY we want to obtain a holistic picture of the role of autophagy in plant-microbe interactions by utilizing bacterial, genetic and cellular diversity. Our central questions are:

  • How does the bacteriome impact autophagy in shoots and roots?
  • Can we use the bacteriome to identify the autophagy degradome and autophagy factors?
  • Does cell-type specific modulation of autophagy influence plant-bacteria interactions?

Within our project, we plan to use a mixture of state-of-the-art approaches including cell biology, proteomics, metabolomics, single-cell transcriptomics and cell-type specific reverse genetic screens.

Requirements:

We are looking for Postdocs (2 positions) and PhD students (f,m,d) with proven expertise in molecular biology. For one of the postdoc position prior experiences with methods to study plant/root-microbe interactions (pathogenic or beneficial), microbial reconstitution assays (SynComs) or microbiome analysis are advantageous. For the other postdoc position expertise in general molecular biology is sufficient. Experience in cell biological approaches (microscopy) is a benefit but not required. Excellent English communication skills as well as teamwork abilities are expected. A master’s degree in molecular biology, biochemistry or corresponding is required for PhD candidates. Work within the different projects/work packages will include microbial, cell biological (confocal microscopy), biochemical (proteomics), genetic (cell-type specific CRISPR-Cas) and single-cell transcriptomic approaches using Arabidopsis and pathogenic/beneficial bacteria as model organisms. No prior experience with autophagy is required!

PhD position in the NanoSignaling Lab.

A PhD position (4 years) is available in our laboratory in the Zentrum für Molekularbiologie der Pflanzen (ZMBP), University of Tübingen.

Our group uses multi-disciplinary approaches in genetics, biochemistry, and live-cell super-resolution microscopy to decipher fundamental aspects of the regulation of cell surface signaling in plants. We notably address how plasma membrane nano-environments are formed and how they orchestrate signaling.

The candidate must hold a master’s degree in biology or biochemistry. Successful candidate will be creative, ambitious, and highly motivated. Previous experience in plant biology and/or live-cell imaging will be an advantage but is not necessarily required. Excellent English written and oral skills, and team-work abilities are required. The specificities of the project will be articulated around the motivations and interests of the candidate. Current research activities relate to cell-surface receptors kinase signaling, salt sensing and evolution.

Plant Molecular Biology

Our research group at the Center for Plant Molecular Biology (ZMBP), University of Tübingen, Germany, is looking for a highly motivated and talented PhD student. The appropriate candidate should have a background in cell and molecular biology and/or biochemistry.

14-3-3 dimers are crucial for the modification of the activity state of various client proteins upon phosphorylation. We currently tackle the analysis of 14-3-3 association with NPH3 – an essential component of the phototropic response (see: bioRxiv preprint doi: https://doi.org/10.1101/2021.04.09.439179). The project aims to address the impact of 14-3-3:NPH3 complex assembly on formation of auxin gradients required for phototropism.

PhD Position in Plant-microbe interaction and cell biology

Research: Our group at the ZMBP Tübingen is interested in the role of the proteasome and autophagy during plant immunity. Recently, we have identified that Pseudomonas activates autophagy for proteasome degradation and enhanced pathogenicity (Üstün et al., 2018). On the contrary, another bacterial pathogen, Xanthomonas campestris pv. vesicatoria, which infects tomato plants, is using a bacterial effector protein to perturb autophagy.

In this project we aim to investigate:

  • Bacterial effectors from Xanthomonas and their impact on autophagy
  • the autophagy pathway in tomato using CRISPR/Cas9 technology
  • Bacterial effectors and their effect on endocytic degradation and vacuolar trafficking

Requirements/Qualifications:

We are looking for a talented PhD student (f,m,d) with proven expertise in cell and molecular biology. Prior experiences with methods to study plant-microbe interactions and advanced confocal microscopy are highly advantageous but not required. Excellent English communication skills as well as teamwork abilities are expected. A master’s degree in molecular biology, biochemistry or corresponding is required. We offer an innovative and scientifically stimulating environment and use a combination of state-of-the-art techniques. The work will include cell biological (confocal microscopy), biochemical (Co-IP, protein purification, protein-protein interactions), genetic and transcriptomic approaches using tomato, tobacco, Arabidopsis and Xanthomonas as model organisms.

Master Thesis Project

“Design and Characterization of a Nanosensor for In Vivo Turgor Measurements in Plant Cells”

The turgor of plant cells is crucial for plant stability and survival. Environmental cues such as drought or altered osmotic pressure lead to the change in turgor pressure having many functional consequences for the plant cell.

Therefore, the monitoring of the turgor pressure in the living plant cell is of great interest However, there is presently no technique to follow turgor changes in real time in living plant cells at cellular resolution.

Based on molecular, biochemical and biophysical considerations we, the research groups of Erik Schäffer and Klaus Harter, developed a concept and strategy for the making of a Förster Resonance Energy Transfer (FRET)-based, genetically encoded nanosensor that should facilitate the measurement of dynamic changes in the plant cell´s turgor.

The thesis work is mainly about

  • Cloning of the nanosensor gene on the basis of already available DNA fragments using the Golden Gate technology
  • Transformation of the nanosensor construct into plant cells and analysis of its expression and intracellular localization by (confocal) light microscopy
  • Determination of the nanosensor´s FRET efficiency under dynamic osmotic stress conditions using spectro-microscopical approaches (e.g. Fluorescence Lifetime Imaging: FLIM)

In addition to biology students with molecular background, the work might also be of interest for master students in Nano-Science and Biochemistry.

Master and or Bachelor Thesis in Plant Pathogen Interactions (RG El Kasmi)

Our research group in the Department for Plant Physiology at the Center for Plant Molecular Biology (ZMBP), University of Tübingen, is offering the possibility for a highly motivated Master and or Bachelor (of Biology, Biochemistry or Cellular Nanoscience) student to do her/his thesis project in our lab.

We are currently working on the characterization of interactors of a plant immune receptor of the NLR family. We would like to validate the preliminary results (interactions observed in yeast-2-hybrid screen) and to further cell biologically, genetically and biochemically characterize the function of the identified proteins during (NLR-mediated) immunity.
The project will include the following methods:
•    Molecular analysis (cloning, bacterial transformation, etc.)
•    In planta Protein-protein interaction studies (co-immunoprecipitation, BiFC and FRET interaction assays)
•    Genetic studies (crosses, genotyping, etc.)
•    Plant infection assays

Gärtnermeister/in, Agrarwissenschaftler/in

oder Biologen/in (m/w/d)

mit einschlägigen theoretischen und praktischen Erfahrungen im Bereich der Pflanzenwissenschaften und –kultur (100 %, E 9 TV-L).

Das Aufgabengebiet umfasst die Organisation und Verantwortung der gesamten Pflanzenanzucht des ZMBP (mehrere Klimakammern, Gewächshäuser und ein Versuchsfeld) einschließlich aller arbeitssicherheitsrelevanten Belange, Führung der Mitarbeiter/innen sowie den technischen Betrieb der Klimakammern, der Gewächshäuser und des Versuchsfelds mit seinen zugehörigen landwirtschaftlichen Maschinen.
