Determination of cell growth

Due to the significance of cell-cell interactions in health and disease there is a keen interest in the development of analytical techniques that enable the analysis of contacts with respect to kinetics, strengths and the underlying signalling processes.

Antibodies directed against the CD3e chain of the TCR/CD3 complex represent a strong T cell stimulus. Contact of T cells with anti-CD3-functionalized surfaces leads to a rapid spreading of cells on the surface within 2-5 minutes.

With RIfS it was possible to detect the stimulus dependent spreading of the T-cell line Jurkat label free and time resolved in flow system. Anti-CD3 antibodies were immobilised onto the RIfS transducers and the spreading of the jurkat cell line as well as the signal reduction of the lck-deficient cell line JCaM 1.6, the use of the spreading inhibitor cytochalsine D and Jurkat cell on different surfaces could be detected.
