Lab Rotation
During a lab rotation a member of the RTG is organizing a lab-training course in a special method, which is established in his lab. This lab course is offered to other RTG students. The visiting students will thereby learn new techniques, experience different levels of expertise and new environments.
Examples for lab rotation are:
October 2018
Yu-Ming Huang (WG Kleindienst) realized protein purification and quantification at the Forchhammer Lab.
December 2017
Moritz Koch (WG Forchhammer) realized GC-analyses at the Jendrossek Lab in order to detect diferent PAH variants in cyanobacteria.
February 2017
Sergii Krysenko (WG Wohlleben) spent some days at the Proteolysis Lab with Prof. Gomis-Rüth (Molecular Biology Institute of Barcelona) in order to learn protein cristallization techniques. Helpful and substantial discussion and tips lead to successful His-GlnA3 purification.
Krysenko has also been in a continuous lab rotation with the WG Brotz-Oesterhelt for protein analytics using the Äkta equipment.
November 2016
- Dmitry Shvarev (WG Maldener) did a three weeks stay at the Descartes Universitsy (Paris) with Dr. Broutin. The aim of this lab rotation was tolearn of protein expression and purification techniques. During his stay different cell strains and culturing conditions, as well as protein extraction and purification methods were tested. Also the way of expression of one subunit of the transporter was established
- Thomas Hagemann (WG Frick) spent some days at the Frochhammer group in order to get familiar with the peristaltic pump, which had to be used fo protein purification. The lab rotation was also helpful to learn the process of equilibrating histidine column and using it for purification. Learning this method
gives the basis to further continue using automatized systems like the Äkta.
August 2016
Thomas Hagemann (WG Frick) went to the Kemijski inštitut. Ljubljana to the lab of Prof. Roman Jerala. The motivation for this lab rotation was to learn how his group obtains this pure E. coli Nissle flagellin without traces of LPS. He learned to manage nickel-NTA columns and to optimize the purification protocol to attain the best outcome in the obtention of highly purified FliC
December 2015
Sergii Krysenko (WG Wohlleben) learned in MPI with Nermin Akduman (WG Sommer) different methods for working with nematodes such as preparation of agar plates for cultivation of C. elegans and P. pacificus, their storage and microscopy examination.
May 2015
- Anna Lange (WG Frick) showed Isabel Hinderberger (WG Mayer) how to handle anaerobic culture chambers for cultivating anaerobic S2 organism Tannerella forsythia.
- Amit Srivastava (WG Forchhammer) cooperated with Klaus Brilisauer in the WG Grond to determine carotenoids from Anabaena cell extracts.
- Alexander Klotz (WG Forchhammer) performed nitrate and nitrite detection using flow-injection analysis (FIA) in the study group Kappler, supervised by Anna-Neva Visser (WG Kappler).
April 2015
- Claudia Laux (WG Peschel) visited Martin Konnerth’s lab (WG Grond) in order to investigate nasal isolates by HR-mass spectrometry.
- Anna Lange went for a lab rotation with Shilpa George (WG Wolz) which was necessary to learn how to generate automated bacterial growth curves with a Tecan reader.
February 2015
Sandra Unsleber (WG Mayer) learned in Klaus Brilisauer (WG Grond/Forchhammer) lab how to use NMR and to understand the software Mnova. She used this for verification of purity and determination of purified amount and chemically isolated peptidoglycan-wall teichoic acid-linker ManNAc(β1→4)GlcNAc.
Summer Term 2014 - Winter Term 2014/2015
Claudia Tominski (WG Geomicrobiology) went to the lab of Marius Spohn and Evi Stegmann (WG Wohlleben) to isolate the Nocardioides sp. from the KS enrichment culture and identify secondary metabolic products produced by the Nocardioides sp. which the iron-oxidizer might need to survive in the enrichment culture. Tominski learned to make different specific and general media for isolation of Actinomyces and to pick and cultivate the isolates.
November 2014
Claudia Tominski (WG Geomicrobiology) did a lab rotation with Sonja Mayer and Regine Stemmler (WG Götz) because she needed a genetic safety level S2 laboratory to order and to cultivate a S2 microorganism for her experiments. She learned how to handle and cultivate a lyophilized culture from the DSMZ and how to store the grown culture at -80°C for further experiments.
October/November 2014
Alexander Zipperer (WG Peschel) went for lab rotation to Martin Konnerth’s lab (WG Grond). There he learned techniques of chemical analysis (Reverse Phase HPLC, MS-MS und NMR) for identification and characterization of bacterial secondary metabolites (antibiotics e.g.).
April 2014
During their lab rotation, Jan Lüddecke (WG Forchhammer) showed Ali Coskun how to use the Isothermal Titraton Calorimetry (ITC). They investigated the binding of purified PII-like protein to different molecules like ATP, ADP, c-di-AMP, Mg2+, etc.
Further on they conducted some enzyme assays using the S. aureus Glutamin synthetase transferase with cell extracts of different S. aureus (mutant) strains.
April 2014
Claudia Tominski (WG Geomicrobiology) went to Freiburg for Nano SIMS Messungen
February 2014
Sabrina Rohrer (WG Wohlleben) learned the Gibson cloning technique in the WG Forchhammer (supervised by Alexander Klotz).
February 2014
Annika Schmidt conducted a lab rotation to help Shilpa George (WG Wolz) to use the Bioreactor for her continuous culture evolution experiments. Mrs. Schmidt gave Mrs. George and her colleague a good introduction to the Bioreactor in her lab and also helped them conduct a trial run.
October 2013
Waldemar Hauf (WG Forchhammer) did lab rotation with the geomicrobiology group (WG Kappler), because they have the equipment and know how to detect small organic acids in supernatants of growth media. Hauf tested Mutants in the Acetyl-CoA metabolism for excretion of acetate and other small organic acids.
November/December 2013
Dorothee Weisbrod (WG Grond) needed help and advice to create a deletion mutant. So she went to Alexander Klotz (WG Forchhammer) for lab rotation. Weisbrod learned how to create a construct, using fusion PCR (Primer design, PCR program for fusion PCR, Polymerase) and thereafter she was able to create a deletion mutant.
May 2013 - present
There is a continuous lab rotation between Marius Spohn (AG Wohlleben) and the GRK-members in WG Forchhammer/Grond regarding exchange of methods and techniques.
May 2013
Alexander Klotz (WG Forchhammer) showed Sonja Mayer (WG Götz) how to perform oxygen consumption measurements with a Clark-type oxygen electrode. The experiment was conducted in the Götz Lab, where it contributed to the understanding of the S. aureus' aerobic respiratory chain.
August 2012
Tina Gauger was trying to establish an anoxic chemostat system. For lab rotation she went to Annika Schmidt (WG Döring) because the group was running two chemostat systems partly anoxically. They were comparing pumps, tubing and handling of the systems. Gauger learned how their partly anoxic chemostat system works and how she could apply this for her anoxic chemostat system.
