Summary
We want to elucidate the interconnected turnover and recycling pathways for peptidoglycan (PG) and wall teichoic acid (WTA) in the Gram-positive model organism B. subtilis. In particular, we will determine the fate of the PG-WTA linker structure and will analyze the role of the murQ gene cluster within this process. LC-MS and radio-assays will be developed to identify cell wall turnover products that accumulate in recycling mutants. Molecular genetic techniques and transcriptional studies with genome microarrays will be applied to investigate the regulation of the murQ cluster as well as the regulatory role of cell wall turnover products.
Zusammenfassung
Wir wollen die verknüpften Abbau- und Recyclingwege für Peptidoglycan (PG) und Wandteichonsäure (WTA) im Gram-positiven Modellorganismus B. subtilis aufklären. Insbesondere werden wir das Schicksal der PG-WTA Verknüpfungsstruktur bestimmen und die Rolle des murQ Genclusters in diesem Prozess analysieren. LC-MS Analysen und Radioaktivitätstest werden entwickelt, um Zellwand-Abbauprodukte zu identifizieren, die in Recyclingmutanten akkumulieren. Molekular-genetische Techniken und Transkriptionsstudien mit der Genom-Mikroarrays werden angewendet, um die Regulierung des murQ Genclusters sowie die regulatorische Funktion von Zellwand-Abbauprodukten zu untersuchen.
