Summary

We aim to resolve the pivotal role of cell-wall amidase AmiC2 for cell-cell communication and multicellular development in Nostoc punctiforme. Therefore, localization studies will be performed and interactions between septum proteins will be studied along with biochemical and structural properties of AmiC2. The presumptive septum-localized connexon, predicted to connect adjacent cells, will be investigated by advanced EM methods. Furthermore, the chemical composition of the murein and the effect of AmiC2 mutation on cellular processes will be analyzed in detail.

Zusammenfassung

Wir möchten die essentielle Rolle der Zellwandamidase AmiC2 für zelluläre Kommunikation und mehrzellige Entwicklung in Nostoc punctiforme studieren. Dazu sollen Lokalisations- und funktionelle Interaktionsstudien zwischen AmiC2 und Septumproteinen durchgeführt werden, biochemische und strukturelle Eigenschaften von AmiC2 sollen aufgeklärt werden. Das hypothetische “Connexon”, das benachbarte Zellen im Filament verbindet, soll mit neuester EM-Technologie untersucht werden. Ebenso soll die chemische Zusammensetzung des Mureins und der Einfluss der AmiC2-Mutation auf zelluläre Prozesse detailliert analysiert werden.
